Aplicación del análisis de rango reescalado R/S para la predicción de genes en el genoma vegetal Acta Agron. (Palmira)

La predicción de genes es en la actualidad uno de los principales desafíos de la genómica. La predicción permite realizar experimentos con alta probabilidad de encontrar genes de interés y comparar regiones de ADN de importancia agronómica entre genomas; además, ayuda a restringir los espacios de búsqueda en las bases de datos. Un procedimiento estadístico con base en el análisis R/S y el coeficiente de Hurst fue desarrollado para caracterizar y predecir genes y los componentes estructurales de estos (exones e intrones) en los genomas eucariotas completos de Arabidopsis thaliana, Oriza sativa y Mus musculus. One of the fungal pathogens that causes more agriculture damage is Botrytis cinerea. Botrytis is a constant threat to crops because the fungus infects a wide range of host species, both native and cultivated. Furthermore, Botrytis persists on plant debris in and on the soil. Some of the most serious diseases caused by Botrytis include gray mold on vegetables and fruits, such as grapes and strawberries. Botrytis also causes secondary soft rot of fruits and vegetables during storage, transit and at the market. A redução da camada de ozônio resulta no aumento da radiação ultravioleta que atinge a superfície terrestre, especialmente a radiação ultravioletaB (UV-B). O aumento da radiação poderá induzir a mudanças estruturais e fisiológicas nas plantas, influenciando no seu crescimento e desenvolvimento. O objetivo deste trabalho foi determinar os efeitos da radiação UV-B ambiente sobre a morfologia das folhas de Arabidopsis thaliana desenvolvidas em condições controladas. Background: Cryopreservation refers to the storage of a living organism at ultra-low-temperature for long-term preservation of plant germplasm. The effect of cryopreservation on the efficiency of exogenous gene genetic transformation and expression level were studied herein. In this work, transgenic Arabidopsis thaliana were successfully conserved in vitro by cryopreservation methods. La masiva cantidad de datos biológicos provenientes de las disciplinas "ómicas" y su aprovechamiento en el mejoramiento genético vegetal requiere de nuevos abordajes teóricos y estadísticos que describan de forma satisfactoria principios generales en los genomas. Expression analysis of cell wall assembly and remodelling-related genes in Arabidopsis roots subjected to boron stress and brassinosteroid at different developmental stages Acta Botanica

ABSTRACT Plant cell walls are affected by many biotic and abiotic stress conditions. The aim of this study is to determine the effects of 24-Epibrassinolide (EBL) on some cell wall-related genes in root tissue of five- and ten-week-old Arabidopsis thaliana plants exposed to boron (B) deficiency (0 µM) or toxicity (3000 µM) at the transcriptional level. Expressions of the genes that encode cellulose synthase (CESA1, CESA4, CESA6 and CESA8), cellulose synthase-like (CSLB5), expansin (EXPA5, EXPA8 and EXPA14) and cell wall protein (SEB1) decreased under conditions of B deficiency and toxicity. We are using molecular, biochemical, and genetic approaches to study the structural and regulatory genes controlling the assimilation of inorganic nitrogen into the amino acids glutamine, glutamate, aspartate and asparagine. These amino acids serve as the principal nitrogen-transport amino acids in most crop and higher plants including Arabidopsis thaliana. We have begun to investigate the regulatory mechanisms controlling nitrogen assimilation into these amino acids in plants using molecular and genetic approaches in Arabidopsis. Retrotransposons (RTEs) are a principal component of most eukaryotic genomes, representing 50%-80% of some grass genomes. RTE sequences have been shown to be preferentially present in disease resistance gene clusters in plants. Arabidopsis thaliana has over 1,600 annotated RTE sequences and 56 of these appear to be expressed because of the exact expressed sequence tag (EST) matches and the presence of intact open reading frames. The objective of this work was to evaluate the effect of the transgenic soybean BR-16-AHAS genetic constitution on the tolerance to the herbicide imazapyr. BR-16-AHAS was crossed with ten other genotypes. The experimental design was a complete randomized block, in a 2x12x3 factorial arrangement, with two sowing periods (winter and summer), twelve crossing groups and three plant positions (upper, mid and lower), with three replicates. The soc1 ful double mutant of A. thaliana produced substantial secondary growth throughout all aboveground stems, whereas in the Col accession only a few cell layers of wood were produced at the base of old stems. The Ca2+ pump inhibitor, thapsigargin, inhibits root gravitropism in Arabidopsis thaliana Biol. Res.

Thapsigargin, a specific inhibitor of most animal intracellular SERCA-type Ca2+ pumps present in the sarcoplasmic/endoplasmic reticulum, was originally isolated from the roots of the Mediterranean plant Thapsia gargancia L. Here, we demonstrate that this root-derived compound is capable of altering root gravitropism in Arabidopsis thaliana. Thapsigargin concentrations as low as 0.1 µM alter root gravitropism whereas under similar conditions cyclopiazonic acid does not. Furthermore, a fluorescently conjugated thapsigargin (BODIPY FL thapsigargin) suggests that target sites for thapsigargin are located in intracellular organelles in the root distal elongation zone and the root cap, regions known to regulate root gravitropism

The impact of mechanical compression on cortical microtubules in Arabidopsis: a quantitative pipeline ArchiMer

Exogenous mechanical perturbations on living tissues are commonly used to investigate whether cell effectors can respond to mechanical cues. However, in most of these experiments, the applied mechanical stress and/or the biological response are described only qualitatively. We developed a quantitative pipeline based on microindentation and image analysis to investigate the impact of a controlled and prolonged compression on microtubule behaviour in the Arabidopsis shoot apical meristem, using microtubule fluorescent marker lines. We found that a compressive stress, in the order of magnitude of turgor pressure, induced apparent microtubule bundling.
